Investigación
El correcto funcionamiento de la fisiología neuronal, y por tanto, de las funciones del sistema nervioso, depende en un alto grado de su polarización morfológica y funcional, que permite una transmisión vectorial de la información entre las dendritas y la terminación axonal. Entre ambos dominios se encuentra el Segmento Inicial del Axón (SIA) con una longitud de entre 20 y 60 micras, siendo el lugar de generación de los potenciales de acción en respuesta a las señales recibidas por la neurona. Ello es posible por la alta concentración de canales iónicos dependientes de voltaje. El segmento inicial del axón es de extrema importancia en la neurona, y la fisiología neuronal depende de su correcta función. Además de ser el lugar donde se genera el impulso nervioso, actúa como una barrera que diferencia los dominios somato-dendrítico y axonal, controlando el tráfico de proteínas hacia el axón. La disfunción o alteración de la composición del SIA está relacionada con múltiples enfermedades del sistema nervioso (p. ej: Síndrome de Angelman, Alzheimer, schizophrenia, trastorno bipolar, autismo, etc..), así como lesiones generadas por daño traumático en el cerebro o por isquemia.

El segmento inicial del axón posee un alto grado de plasticidad, pudiendo cambiar su distancia al soma, su longitud, así como la concentración de canales iónicos. Estos cambios permiten adaptar la amplitud y número de potenciales de acción en función de la señal recibida por las neuronas en condiciones fisiológicas o patológicas. Un mayor conocimiento de la composición y mecanismos regulatorios del SIA podría generar estrategias terapéuticas que mantengan la supervivencia neuronal y controlen cambios en la excitabilidad neuronal asociados a enfermedades y trastornos del sistema nervioso.
